From a young age, Calamaro showed an early inclination for music, receiving his first instrument, an accordion, at the age of eight. Shortly after, he composed his first musical piece. Starting at age thirteen, he dedicated himself to the electric guitar and the piano, learning the latter with Osvaldo Calo. His professional beginnings led him to record his first album in Uruguay as a keyboardist for the group Raíces, recommended by Sergio Makaroff. His official debut as a musician was with this group. Later, he formed Chorizo Colorado Blues Band with Augusto Gringui Herrera, and later explored projects such as Elmer's Band, in which Eduardo Cano also participated. Calamaro also worked as a session musician for artists like Los Hermanos Makaroff and Julián Petrina, and was invited by Héctor Zeta Bosio to join The Morgan, a band that also featured Gustavo Cerati. Subsequently, he was part of Stress and Proyecto Erekto, groups that laid the foundations for Los Estereotipos.
